![]() ![]() Furthermore, we showed that our approach can separately detect and quantify the arrival of membrane proteins at the apical and basolateral plasma membrane in single polarized epithelial cells by detecting the luminescence signals with a microscope, thus opening novel avenues for characterizing the variations in trafficking in individual epithelial cells. ![]() The best results were achieved with split Nanoluciferase, for which luminescence increased more than 6000-fold upon recombination. We compared the performance of split Gaussia luciferase and split Nanoluciferase by using a system to synchronize biosynthetic trafficking with conditional aggregation domains. Once the protein of interest arrives at the cell surface, the luciferase fragments complement and generate luminescence. Here, we introduce a novel approach based on split luciferases, which uses one luciferase fragment as tag on the protein of interest and the second fragment as supplement to the extracellular medium. However, the sensitively measuring the cell surface concentration of a particular protein of interest in live cells and in real time represents a considerable challenge. In particular, epithelial cells tightly control the number of carriers, transporters and cell adhesion proteins at their plasma membrane. (After the topic has moved, I could not edit my first post. Each cell in a multicellular organism permanently adjusts the concentration of its cell surface proteins. You only have to change the Configuration part. INCLUDE_PATTERN = "\.($"ĬOMMAND = "curl -data '' # start of the script WAIT_SEC = 60 # End of configuration # config variables (DO NOT CHANGE!) The wait time is also optional or can be de-/increased.Ĭreate triggered task for startup in Synology which is calling this script. So if you don´t need it, you can remove the part with the PATTERN or change the regex (I´m not a regex expert, but it works like expected.). The script can be easier, because I wanted to exclude files, which only have numbers. # wait for other changes for not re-trigger during the sleep timeĮcho Trigger scan in $WAIT_SEC seconds > $LOGFILEĮcho Trigger library refresh > $LOGFILEĮcho Stop monitoring media files > $LOGFILE # adjust the IP and the api-key to your settings!Įcho Start monitoring media files > $LOGFILEĮcho " The file '$file' appeared in directory '$directory' via '$action'" > $LOGFILE # add additional file extension here and to -include pattern! # NOTE: the PATTERN does not work in -include param (don´t know why.)Įcho Monitoring media files canceled > $LOGFILE # check for mkv and ts files which have at least a character or a bracket So I found another solution/workaround for this problem: a script running on my Synology NAS, which triggers a library refresh task, if a file has created, changed or removed in my media folder: I searched for a solution mainly in this forum, but no luck. I tried to mount my Synology drive with SMB and NFS, but no success. ![]() On Synology I had no problems, but with the mounted shares on my Raspberry I did not get it to work. After moving my Emby Server from Synology Diskstation to Raspberry Pi with DietPI, the real time monitor on my mounted network shares does not work. ![]()
